Pro-inflammatory peptides such as the chemoattractant interleukin-8 are potent activators of the protease-activated receptors (PARs) which are involved in cardiovascular diseases. We have recently demonstrated that interleukin-8 up-regulates the expression of the mRNA of PAR-1, PAR-2 and PAR-3 as well as the expression of the receptor protein. In vascular smooth muscle cells, PAR-2 seems to mediate the increase in [Ca2+]i and the activation of the RhoA/ROCK pathway. The PAR-1 and PAR-3 may be involved in the proliferation of vascular smooth muscle cells. The expression of PAR-2 and PAR-3 is also enhanced in human aortic aneurysms, and PAR-2 mediates the effects of interleukin-8 on smooth muscle cell proliferation.
